Yes, a direct sequel to Skylanders: Spyro's Adventure has been confirmed in the recently revealed Skylanders: Giants!
“Skylanders Giants” will be released this fall, featuring a new game and eight new toys. These figurines will be more than twice the size of the original ones and will cost more, though Hirshberg declined to reveal the price.
Accessories from the first “Skylanders” game will work on the new one. Hirshberg said the sequel might be even more profitable because many buyers will already own the Portal of Power, which was the most expensive part to make for the first game.
It's also been confirmed that all of the current Skylanders will work just fine with the sequel, so you'll have plenty of time to stock up and prepare to continue your adventures.

Two of the eight new normal-sized Skylanders had been revealed, one is called Alchemist, under the Magic element, while the other is an eagle-like Air Skylander.

There will also be remodeled of some of the Skylanders we already know, or will be released. Cynder and Zook are the only ones who have remodeled figurines so far:

Here is the revealed story of the Giants:
"As the story goes, the original Skylanders were actually giants. And it was them who first defeated Kaos and his machines many moons ago. As a form of sacrifice, the Giants were shot to Earth where they’ve been buried underground ever since. The Skylanders will need to summon the Giants back to Skylands to help them fight Kaos and his war machines."
Hans Zimmer will return to compose the sequel's main theme, and Lorne Balfe will once again be composing the soundtrack!
The Toy Story Writers, Joel Cohen and Alec Sokolow, have also returned to help write the story.
In the brand new Starter Pack, you will get three Skylanders (a Giant, a new Skylander, and a returning Skylander), a Portal of Power, Trading Cards, a Poster, and the game itself. The Starter Pack will also be available without the Portal of Power for those who already have it.
EDIT:
We have received confirmation that there will be NO PC version of Skylanders: Giants.
And also: There will be more than 15 levels in the game that feature unique terrain, fortresses, obstacles and other mysteries.
